Biochemistry of the Sphingolipids

چکیده
Hydrolysis of phytoglycolipid by barium hydroxide yielded the various products possible from cleavage of the phosphate diester bond (ceramide phosphate plus oligosaccharide and ceramide plus oligosaccharide phosphate). About 50% of the oligosaccharide was recovered as a phosphorus-free, amorphous solid, the analytical data for which suggested a hexasaccharide structure. Studies on the fractionation of this mixture will be presented in a subsequent publication. It is the purpose of this report to present degradation studies on the crude oligosaccharide which have provided substantial additional evidence as to the basic structures involved. In a previous communication (3), it was shown that crude oligosaccharide on mild acid degradation yields a relatively stable trisaccharide containing inositol, glucosamine, and hexuranic acid. This material is obtained in excellent yields as a crystalline product which accounts for essentially all of the glucosamine and hexuronic acid of the oligosaccharides. Since this unit constitutes a common moiety present in PGLr from a

Sphingolipids: important players in multiple sclerosis.
Multiple Sclerosis (MS) is the most common cause for permanent disability in young adults. Current pathophysiological understanding has identified an autoaggressive immune reaction with infiltration of immune cells into the central nervous system and local inflammatory and demyelinating reactions. The current therapy focuses on a modulation or suppression of immune functions. Sphingolipids, mai...
متن کاملSphingosine kinase: biochemical and cellular regulation and role in disease.
Sphingolipids have emerged as molecules whose metabolism is regulated leading to generation of bioactive products including ceramide, sphingosine, and sphingosine-1-phosphate. The balance between cellular levels of these bioactive products is increasingly recognized to be critical to cell regulation; whereby, ceramide and sphingosine cause apoptosis and growth arrest phenotypes, and sphingosine...
متن کاملThe control of the balance between ceramide and sphingosine-1-phosphate by sphingosine kinase: oxidative stress and the seesaw of cell survival and death.
Sphingolipids are components of all eukaryotic cells that play important roles in a wide variety of biological processes. Ceramides and sphingosine-1-phosphate (S1P) are signaling molecules that regulate cell fate decisions in a wide array of species including yeast, plants, vertebrates, and invertebrates. Ceramides favor anti-proliferative and cell death pathways such as senescence and apoptos...
متن کاملThe role of sphingomyelin breakdown in measles virus immunmodulation.
Measles virus (MV) efficiently causes generalized immunosuppression which accounts to a major extent for cases of measles-asscociated severe morbidity and mortality. MV infections alter many functions of antigen presenting cells (APC) (dendritic cells (DCs)) and lymphocytes, yet many molecular targets of the virus remain poorly defined. Cellular interactions and effector functions of DCs and ly...
متن کاملThe complex life of simple sphingolipids.
The extensive diversity of membrane lipids is rarely appreciated by cell and molecular biologists. Although most researchers are familiar with the three main classes of lipids in animal cell membranes, few realize the enormous combinatorial structural diversity that exists within each lipid class, a diversity that enables functional specialization of lipids. In this brief review, we focus on on...
متن کاملBacterial membrane lipids: diversity in structures and pathways.
For many decades, Escherichia coli was the main model organism for the study of bacterial membrane lipids. The results obtained served as a blueprint for membrane lipid biochemistry, but it is clear now that there is no such thing as a typical bacterial membrane lipid composition. Different bacterial species display different membrane compositions and even the membrane composition of cells belo...
